Item Avaliação do fluxo sanguíneo do folículo ovulatório e corpo lúteo de fêmeas bubalinas por ultrassonografia Doppler(2024-09-27) Farias Júnior, Francisco Timbó; Batista, André Mariano; http://lattes.cnpq.br/5615914349535394; http://lattes.cnpq.br/7562129132084802The Mandatory Supervised Internship (ESO) is an essential part of the training of a Veterinary Doctor, being a mandatory requirement in the Veterinary Medicine course at UFRPE. With a workload of 420 hours, the internship was conducted at the Laboratory of Biotechniques Applied to Reproduction (LBR), between 04/01/2024 and 06/14/2024. The focus was on improving reproductive biotechnology, with emphasis on the in vitro maturation of oocytes and the development of an article based on research carried out with Murrah buffaloes. The study evaluated the blood flow of the ovulatory follicle and corpus luteum using Doppler ultrasound, correlating these data with reproductive efficiency. In the laboratory, activities such as in vitro maturation of bovine oocytes (IVM) and evaluation of meiotic progression using epifluorescence microscopy were carried out. Under the guidance of Professor Dr. André Mariano Batista and supervision of Professor Dr. Cláudio Coutinho Bartolomeu, the internship provided a practical insights into reproductive biotechnologies and their impact on reproductive efficiency in buffaloes.Item Relatório de Estágio Supervisionado Obrigatório (ESO) realizado na Empresa Brasileira de Pesquisa Agropecuária - EMBRAPA Gado de Leite, Juiz de Fora e no Laboratório de Biotécnicas Aplicadas à Reprodução do Departamento de Medicina Veterinária da Universidade Federal Rural de Pernambuco: efeito da adição de nanoemulsão à base de óleo essencial rico em limoneno ao diluente de refrigeração de sêmen caprino(2024-02-15) Nottar, Luiz Henrique Todescatt; Batista, André Mariano; http://lattes.cnpq.br/5615914349535394; http://lattes.cnpq.br/3747311074315408The present study aims to report the activities carried out in compliance with the discipline 08525 - Mandatory Supervised Internship (MSI), part of the Bachelor's Degree in Veterinary Medicine, conducted between October 2, 2023, and November 28, 2023, at the Brazilian Agricultural Research Corporation (Embrapa), in the Dairy Cattle unit in the municipality of Juiz de Fora - MG, and between January 2 and January 19, 2024, at the Laboratory of Biotechniques Applied to Reproduction (LBR) of the Department of Veterinary Medicine (DMV) of the Federal Rural University of Pernambuco (UFRPE). The activities carried out provided in-depth training in in vitro embryo production (IVP) techniques for research purposes, including follicular aspiration of ovaries from abattoirs, in vitro maturation (IVM), in vitro fertilization (IVF), and in vitro embryo culture (IVC). Activities in the field of molecular biology involving DNA extraction and amplification through the Polymerase Chain Reaction (PCR) technique were also performed. In the second chapter of this study, a research was conducted to evaluate the effect of adding nanoemulsion based on limonene-rich essential oil extracted from wild orange peel (NELS) to the semen extender of goat breeders. Seminal samples from five Saanen breed goats were used for the research. Each seminal pool was diluted in Tris-egg yolk supplemented with NELS at concentrations of 1.25, 2.5, and 5% at a concentration of 200 × 106 sperm/mL. The samples were placed in Eppendorf tubes (1.5 mL) and stored in refrigeration boxes (botuflex®) for cooling curve determination up to 5 ºC, and after 2 hours, they were transferred to the refrigerator, also at a controlled temperature of 5 ºC. For each analysis, part of each sample was aliquoted and transferred to other Eppendorf tubes (1.5 mL) in a dry bath at 37 ºC and subsequently evaluated in the Computer-Assisted Sperm Analysis System (CASA) for sperm kinetics, and in epifluorescence microscopy for Mitochondrial Membrane Potential (MMP), Plasma Membrane Integrity (PMI), and Acrosomal Membrane Integrity (AMI). The addition of wild orange nanoemulsion to the Tris-egg yolk extender was not able to improve sperm motility and kinetic parameters, besides significantly reducing MMP after 24 hours of refrigeration.Item Relatório de Estágio Supervisionado Obrigatório, realizado na Clínica Univet Saúde Animal em Santa Cruz do Capibaribe - PE. In horses, it was only used in the medical-clinical area. The biggest focus was on bovine reproduction with fixed-time artificial insemination (IATF). During the internship period, farms in the forest and rural areas of Pernambuco were visited, mainly working with beef cattle. Gestational diagnoses were made with several different age ranges, protocol initiations, and artificial inseminations. In the surgical clinic area, surgeries such as castration in horses, vulvoplasty in heifers, and hematoma drainage in calves were experienced. Laboratory and field tests were carried out, with laboratory tests for brucellosis using the AAT (buffered acidified antigen) method and field tests focused on tuberculosis. At the animal exhibition, it was possible to receive documents (GTA, brucellosis test, equine infectious anemia test, and vaccination card) to receive the animals that arrived at the Cordeiro exhibition park. The birth of a Girolando female was observed. Chapter II included a literature review with the title “Use of equine chorionic gonadotropin to improve TAI results in Nelore females” in which the work reaffirms the effectiveness of using eCG in postpartum cows with low body scores, helping TAI protocols as a tool to enhance and improve follicular development and maturation.Item Comparação entre quatro protocolos de preparação do plasma rico em plaquetas na espécie equina(2024-02-02) Xavier, Gabriela Reis; Carneiro, Gustavo Ferrer; http://lattes.cnpq.br/0903050581306709; http://lattes.cnpq.br/3490771668260807The presente work aims to describe the activities carried out during the compulsory supervised internship, a compulsory subject for completion of the Veterinary Medicine course at the Federal Rural University of Pernambuco. The internship was initially carried out at the Fazenda Sucupira experimental field - Embrapa Recursos genéticos e Biotecnologias, Km 5 of DF 001, Riacho Fundo II and was then completed at the Animal Andrology Laboratory, in the Department of Veterinary Medicine at the Federal Rural University of Pernambuco (ANDROLAB/DMV/UFRPE), under the guidance of Prof. Dr. Gustavo Ferrer Carneiro during the period from 02/11/2023 to 16/01/2024. Throughout the internship, it was possible to participate in the routine of Fazenda Sucupira and the ANDROLAB routine, accompanying and assisting in carrying out the equine reproduction experiment, with an emphasis on follicular control, artificial insemination in horses, equine embryo lavage, semen collection and freezing. It was also possible to follow other research activities carried out at the internship site and with other species. Ovariectomia unilateral em égua com infertilidade idiopática - Relato de caso(2023-09-18) Pereira, Eliana Nunes; Carneiro, Gustavo Ferrer; http://lattes.cnpq.br/0903050581306709; http://lattes.cnpq.br/5484384582010620The Compulsory Supervised Internship (ESO), a compulsory subject in the last term of the Veterinary Medicine course, took place in two stages: the first was in the field, in the area of equine reproduction, with Dr. Sandra Victalino at 6 properties in the municipality of Moreno, located in the Metropolitan Region of Recife-PE, and in the municipalities of Chã Grande and Gravatá, both located in the Central Agreste region of Pernambuco, between the dates of 06/05/2023 and 07/21/2023. The second stage of the internship took place at the Large Animal Outpatient Clinic (AGA) of the Department of Veterinary Medicine (DMV) of the Federal Rural University of Pernambuco (UFRPE), in the area of clinic, surgery and reproduction, from 07/24/2023 to 08/30/2023, from Monday to Friday, accounting for a total of 420 hours of supervised internship. During the ESO period, students were supervised by Prof. Dr. Gustavo Ferrer Carneiro, professor at UFRPE, and by two supervisors, one for each period: Dr. Sandra Lacet Victalino Mancebo and Prof. Dr. Sandra Regina Fonseca de Araújo Valença, professor at UFRPE. The ESO period allows students to have more autonomy in directing their internship towards the area in which they feel the most affinity or even use this period to try out new areas that they did not have the opportunity to during their undergraduate studies. In this way, the aim of this work is to report on the activities carried out during the ESO period, as well as to report on a case monitored during this period concerning a mare, who is currently 5 years old and who, since she reached reproductive age, has been unsuccessful in her reproductive activity After a few years of assessment and detailed hormonal testing and imaging, the mare presented an hypertrophy of right ovary with a suspicion of a granulosa cell tumor. As a result of this suspicion and the fact that the donor mare was unable to cycle, the decision was made to perform a unilateral ovariectomy in an attempt to return the contralateral ovary to normal hormonal activity. After a histopathological examination, the ovary showed a neoplasm of the sex cord, suggestive of Tecoma. The internship was carried out in the Animal Reproduction Area of the Department of Veterinary Medicine of the Federal Rural University of Pernambuco, located on Av. Dom Manoel de Medeiros S/N, in Dois Irmãos, Recife-PE. Under the guidance of Prof. Dr. Gustavo Ferrer Carneiro and supervision of Prof. Dr. Maria Madalena Pessoa Guerra during the period from 05/29/2023 to 08/11/2023. Throughout the internship it was possible to participate in the routine of the Animal Andrology laboratory, accompanying and assisting in the performance of andrological examinations; semen collection and evaluation; Conducting tests to determine the freezing capacity of semen, evaluation of sperm morphology and reproductive potential aiming at final evaluation of the reproductive potential of the animals evaluated. In addition, it was also possible to follow the research activities developed at the internship site. Francisco da Silva Alves Pinheiro, in the Metropolitan, Zona da Mata and Agreste region of the State of Pernambuco, where clinical and surgical procedures, nutritional management, blood collection for laboratory tests, weighing and vaccination of the animals of the herd, examinations were followed. ultrasound and reproductive management of cattle. Most of the internship hour load was aimed at monitoring the protocol steps and performing fixed-time artificial insemination (FTAI) in females, as well as inducing puberty in heifers and pregnancy diagnoses. This paper covers a description of the internship location, its physical and administrative structures, its daily routines and activities resulting in a literature review.
