Item Relatório de Estágio Supervisionado Obrigatório (ESO) realizado na Empresa Brasileira de Pesquisa Agropecuária - EMBRAPA Gado de Leite, Juiz de Fora e no Laboratório de Biotécnicas Aplicadas à Reprodução do Departamento de Medicina Veterinária da Universidade Federal Rural de Pernambuco: efeito da adição de nanoemulsão à base de óleo essencial rico em limoneno ao diluente de refrigeração de sêmen caprino(2024-02-15) Nottar, Luiz Henrique Todescatt; Batista, André Mariano; http://lattes.cnpq.br/5615914349535394; http://lattes.cnpq.br/3747311074315408The present study aims to report the activities carried out in compliance with the discipline 08525 - Mandatory Supervised Internship (MSI), part of the Bachelor's Degree in Veterinary Medicine, conducted between October 2, 2023, and November 28, 2023, at the Brazilian Agricultural Research Corporation (Embrapa), in the Dairy Cattle unit in the municipality of Juiz de Fora - MG, and between January 2 and January 19, 2024, at the Laboratory of Biotechniques Applied to Reproduction (LBR) of the Department of Veterinary Medicine (DMV) of the Federal Rural University of Pernambuco (UFRPE). The activities carried out provided in-depth training in in vitro embryo production (IVP) techniques for research purposes, including follicular aspiration of ovaries from abattoirs, in vitro maturation (IVM), in vitro fertilization (IVF), and in vitro embryo culture (IVC). Activities in the field of molecular biology involving DNA extraction and amplification through the Polymerase Chain Reaction (PCR) technique were also performed. In the second chapter of this study, a research was conducted to evaluate the effect of adding nanoemulsion based on limonene-rich essential oil extracted from wild orange peel (NELS) to the semen extender of goat breeders. Seminal samples from five Saanen breed goats were used for the research. Each seminal pool was diluted in Tris-egg yolk supplemented with NELS at concentrations of 1.25, 2.5, and 5% at a concentration of 200 × 106 sperm/mL. The samples were placed in Eppendorf tubes (1.5 mL) and stored in refrigeration boxes (botuflex®) for cooling curve determination up to 5 ºC, and after 2 hours, they were transferred to the refrigerator, also at a controlled temperature of 5 ºC. For each analysis, part of each sample was aliquoted and transferred to other Eppendorf tubes (1.5 mL) in a dry bath at 37 ºC and subsequently evaluated in the Computer-Assisted Sperm Analysis System (CASA) for sperm kinetics, and in epifluorescence microscopy for Mitochondrial Membrane Potential (MMP), Plasma Membrane Integrity (PMI), and Acrosomal Membrane Integrity (AMI). The addition of wild orange nanoemulsion to the Tris-egg yolk extender was not able to improve sperm motility and kinetic parameters, besides significantly reducing MMP after 24 hours of refrigeration.Item Acompanhamento e controle zootécnico do sistema de produção de pequenos ruminantes (caprinos e ovinos), do CODAI/UFRPE, localizado na região da Zona da Mata Norte, no município de São Lourenço da Mata - PE(2021-12-07) Brito, Admilson Ribeiro Toscano de; Freitas, João Antônio Gomes de; http://lattes.cnpq.br/1765326853404114; http://lattes.cnpq.br/7228033819346383The Obligatory Supervised Internship (ESO) was held at College Agricole Dom Agostinho Ikas of UFRPE, Campus-Tiúma, in the Animal Science Area, in the Nucleus de Caprinovinocultura, located in the forest Mata Norte region, in the district of Tiúma, municipality of San Lourenço of the Mata, Pernambuco, under the supervision of teacher Dr. João Antônio Gomes de Freitas and supervision of teacher Drª. Elisa Alves Vilar, with a total workload of 420 hours, in the period from 08/30/2021 to 11/16/2021. Zootechnical Monitoring and Control of the Goats and Sheep Production System were carried out, involving management of facilities; food management; reproductive management; productive management; sanitary management and disposal management. This work aimed to report the main activities related to the experiences developed during the referred internship, in the goat and sheep production nucleus of CODAI-UFRPE, in the routine of raising goats and sheep. The production nucleus, mentioned above, has a fixed squad of 22 animals; being 10 goats and 10 sheep (parents); 01 goat and 01 ram (breeders), in addition to the offspring, calving, which occur during the breeding routine. The goats are predominantly of the Anglo-Nubiana breed, while the sheep come from the Santa Ines breed. The intensive production system, with an emphasis on animal welfare, is the method adopted by the production nucleus. The installations consist of suspended bays with slatted floors. Nutritional and food management is carried out in a balanced way with elephant grass pasture (Pennisetum purpureum) and supplemented with protein and mineral concentrate. The main diseases observed and treated during this period were some bacterial diseases such as caseous lymphadenitis, food poisoning, epidermatitis and parasitic diseases such as worms. Discards are carried out following the guidelines for the genetic and productive improvement of the herd. During this period the animals showed an average weekly weight gain ranging from 250g to 350g per animal (50g/day).Item Relatório do Estágio Supervisionado Obrigatório (ESO), realizado no Laboratório de Andrologia e Ambulatório de Grandes Animais do Departamento de Medicina Veterinária da Universidade Federal Rural de Pernambuco(2018-08-31) Lima, Carlos André Ferreira; Bartolomeu, Cláudio Coutinho; http://lattes.cnpq.br/7619056086707716; http://lattes.cnpq.br/5611281478872633The Mandatory Supervised Internship (ESO) was conducted under the guidance of Prof. Dr. Cláudio Coutinho Bartolomeu. The first moment was experienced in the premises of Andrology Laboratory - Androlab in the Department of Veterinary Medicine of UFRPE, under the supervision of Professor Dr. André Mariano Batista. The second one was carried out at the Ambulatory of Large Animals of the Veterinary Hospital of UFRPE (AGA-HOVET / UFRPE), under the supervision of Professor Dr. Huber Rizzo. At ANDROLAB the internship was carried out in the period from June 1st to July 13th, 2018, totaling 246 hours. The second part held at AGA-HOVET / UFRPE comprised the period from July 27 th to August 24 th, 2018, totaling 174 hours. This report aims to describe the activities carried out in the area of andrology and reproduction biotechnologies, comprising semen collection of goats and sheep, macro and microscopic analysis, cryopreservation and thawing. In the area of veterinary medical practice of horses and ruminants, comprising clinical examination of hospitalized animals, administering the prescribed drugs and helping to perform complementary tests when necessary. From the experiences obtained, a clinical case called attention: anomaly of the development of the reproductive apparatus in goats. It is an infant patient, attended during the internship. It presented important anatomical changes in the external genital organs, making it difficult to determine the sex of the animal.
